Seminal Fluid Analysis (SFA) Semen:

Semen:
Which is ejaculated during the male sexual act. Is composed of the fluid and sperm from the vasdefrense. Fluid (from seminal vesicle 60%) (from prostate gland 30%) and small amounts from the mucous gland. Specially bulbourethral gland. We can divided the SFA to two steps:
A- Macroscopic examination.
B- Microscopic examination.
A- Macroscopic examination:
1) Appearance:
The semen samples were examined immediately after liquefaction or with in 1 hr. of ejaculation at room temperature (normal semen samples have homogenous Grey-opalescent appearance).

3) Abstinence period:
The time of prevention or discontinue from coitus. Range from 2-7 days.
4) Liquefaction time:
normal semen samples liquefied within 20-30 min. At CO2 incubator while 1 hr. at room temperature.

5) Viscosity:
it was estimated by pasture pipette or syringe with out needle and then allowing the semen to drop by gravity and observing the length of the thread normal samples leaves the pipette as small discreet drops after liquefaction time. The normal viscosity while form less than 6 cm.
Note:
The semen samples viscous immediately after ejaculation due to seminal vessels secretion (Coagulum protein) and then will liquefied due to prostate secretion
(Profibrinolysin enzyme).
