Non catecholamine :
*Phenylephrine :
-selective ?1 agonist
-vasoconstrictor so it increase systolic and diastolic BP
Uses:
-topically in mm congestion
-ophthalmic solution for mydriasis
-used to increase BP
SE:
large dose cause hypertensive headache and cardiac irregularity
*methoxamine
-selective ?1 agonist
-vasoconstrictor and increase BP
-used to relieve supra-ventricular tachycardia and to over come
hypotension during surgery with halothane anesthetic
* clonidine
-selective ?2 agonist
-used to decrease BP
-used in with drawal from opiate BDZ
* Terbutaline & Albuterol
- ?2 agonist
-Short acting & rapid onset
-bronchodilator
-Relax uterus( used in premature labour)
* Metaprotrenol
- ? agonist (?2> ?1)
-dilate bronchioles
-Used in asthma
*Salmeterol
- selective ?2 agonist
-Long acting & slow onset
-bronchodilator
*Salbutamol
- selective ?2 agonist
-Used in asthma , used in premature labor and prevent abortion.
*Ritodrine
- selective ?2 agonist
-used for dely labor
Indirect acting adrenergic agonist :
These drug cause nor adrenaline release from presynaptic terminals & (
not bind with R ) potentiate the effect of endogenously produced nor
adrenaline.
*Amphetamine
- ? , ? effect and CNS.
-centrally stimulatory action ( cause euphoria ).
-used orally , iv , infusion , smoking.
-increase BP (? )
-stimulate heart (? )
-increase motor activity
- increase self confidence
- stimulate respiration
Uses:
-hyperactive child
-narcolepsy
-decrease appetite
CI: in pregnency
Mixed action adrenergic agonist:
They induce release of nor adr. And activate adrenergic R
*Ephedrine:
-An alkaloid
-has ?1 , ? effect
-its effect is slowly but longer than adr.
-penetrate CNS
-vasoconstriction & cardiac stimulation( increase systolic and diastolic
BP)
-used as mydriatic agent
-bronchodilator( in prophylaxis of asthma)
-treatment of MG (it enhance cholinergic NM transmission)
- in adult cause alertness , anxiety , insomnia , tremor while in child cause
sleep
-nasal decongestant
-used to increase BP
*Norephedrine: same with CNS effect
